Abstract
This study develops sirolimus formulations based on polymeric micelles for the topical treatment of facial angiofibromas in tuberous sclerosis complex. Using TPGS micelles and the cutaneous biodistribution method, we designed stable aqueous micellar solutions and hydrogels with improved sirolimus incorporation and favorable release properties. Experiments using porcine skin confirmed higher sirolimus delivery into the viable epidermis compared with the commercial ointment. These results show that polymeric micelles provide an effective platform for targeted epidermal delivery and offer a promising non-invasive treatment option for facial angiofibromas.
